**L-Carnosine** is the dipeptide **β-alanyl-L-histidine**, concentrated in skeletal muscle and brain, and taken in the Institute's stack as an **anti-glycation and antioxidant** agent — specifically to protect **BH4** from oxidative destruction.

## What it does

Carnosine has four reasonably well-established activities:

- **Anti-glycation** — it reacts with reactive carbonyls and blocks the formation of **advanced glycation end-products (AGEs)**, the irreversible sugar–protein cross-links that accumulate over a lifetime and stiffen tissue.
- **Carbonyl scavenging** — it quenches reactive aldehydes such as 4-HNE and malondialdehyde.
- **Metal chelation** — it binds copper and zinc, limiting metal-catalysed radical formation.
- **pH buffering** — in muscle it buffers the protons generated during high-intensity work, which is the basis of the sports-nutrition use.

## Why it is in a folate stack

L-methylfolate exists in the stack to regenerate **BH4**. But BH4 is lost to **oxidation** as fast as it is lost to use, and oxidised BH4 does not merely fail to work — it causes the NOS enzyme to uncouple and produce superoxide.

So the stack does not only supply the cofactor, it **defends** it. Carnosine occupies the **degradation inhibitor** row of The Amplification Framework — the same functional slot that vitamin C fills on the monoamine side and that galantamine fills in the cholinergic stack. It is the item that stops the amplifier draining.

## The bioavailability problem

**Oral carnosine is largely destroyed before it reaches muscle.** Serum **carnosinase (CN1)** hydrolyses it rapidly into β-alanine and histidine, and carnosinase activity varies substantially between individuals by genotype.

The practical consequence is well established in sports nutrition: **β-alanine, not carnosine, is the supplement that reliably raises muscle carnosine**, because β-alanine is the rate-limiting precursor and survives to be re-synthesised into carnosine inside the cell. A gram of oral carnosine is, for muscle purposes, an inefficient way of taking β-alanine.

This does not void the systemic argument — intact carnosine in plasma does scavenge carbonyls during its short residence, and the brain synthesises its own — but it is the same recurring lesson: **the form is the intervention.**

## Life extension

Carnosine extended median lifespan in **senescence-accelerated mice**, and the effect reported in combination with **vitamin E** is the result the Institute's paper cites. These are animal results in a strain selected for accelerated ageing. There is no human lifespan data.

## Cautions

- **β-alanine paraesthesia** — the harmless but genuinely unpleasant skin tingling that follows β-alanine, and that oral carnosine can produce once hydrolysed.
- **Zinc and copper** — carnosine chelates both; relevant at sustained high doses.
- **Hypotension** — mild lowering reported.
- **Pregnancy** — insufficient data.
